Sudhir Kasturi

... to Sudhir Kasturi for converting his 2016 NIH K01 to his first R01 funding this month.

Two of the key obstacles to developing an effective HIV-1 vaccine are: 1) identifying adjuvants and immunogens that induce a high magnitude of persistent, specific antibody responses and 2) overcoming challenges with scale-up manufacturability.

Sudhir's new project -- Novel nanoparticulate adjuvants to enhance HIV-1 Env specific mucosal antibody responses (R01AI145640) -- will address both of these obstacles by using a rhesus macaque model to systematically test novel HIV-1 vaccines that include new adjuvants and emerging HIV-1 envelope protein immunogens in collaboration with the Infectious Disease Research Institute (IDRI) and 3M Pharmaceuticals, who have experience in developing clinical grade vaccines. Their work, if successful, should rapidly inform selection of a HIV-1 vaccine candidates for use in humans.

      Cheryl Day

Cheryl Day and Grace John-Stewart (University of Washington) received new R01 funding to look at The Effect of HIV Exposue and Infection on Immunity to TB in Children (1R01AI142647-01).

TB-related morbidity and mortality is high in HIV-infected children and HIV-exposed uninfected (HEU) children are also at substantial risk of Mtb infection and TB disease.

Anti-mycobacterial innate immune responses are poorly defined in BCG-immunized HIV-infected and HEU infants and may modify TB susceptibility. Cheryl's study will utilize specimens from unique cohorts of HIV-infected, HEU, and HIV-unexposed (HUU) children in Kenya to determine predictors of anti-mycobacterial immunity (including HIV exposure, HIV infection, antiretroviral treatment, and other factors), the influence of HIV exposure or infection on 'trained immunity', and the effects of this immunity on susceptibility to TB.
