The current standard of care for PrEP is a daily oral regimen of tenofovir/emtricitabine (Truvada) but an interim analysis of HPTN 084 data (which enrolled 3,223 cisgender women in seven sub-Saharan African nations) has demonstrated that an every-other-month injection of long acting cabotegravir (CAB LA) was 89% more effective in preventing HIV infection.

The HPTN 083 clinical trial, released earlier this year, showed that CAB LA injected once every eight weeks was also superior to daily oral FTC/TDF for HIV prevention among cisgender men and transgender women who have sex with men.

Mirko Paiardini and MPI Jacob Estes (Oregon Health and Science University) have received new R01 funding (R01MH125457) for use in DETERMINING THE RELATIVE CONTRIBUTION OF CD4 T CELLS AND MACROPHAGES TO HIV PERSISTENCE AND REBOUND.

In this project Mirko and Jacob will lead a multidisciplinary team of researchers in addressing -- using a Rhesus Macaque in vivo model -- how the size and relative distribution of the HIV reservoir in CD4 T cells and macrophages govern viral persistence on ART, the extent of residual inflammation and neuropathogenesis, and viral rebound after ART interruption. The team hope their studies will ultimately inform the design of novel therapeutic strategies aimed at achieving HIV remission.
